    Background:

    • Timoptic (timolol maleate) is a topical beta-blocker used to treat glaucoma.
    • Its effects on corneal reepithelialization are not fully understood.
    • Understanding drug effects on corneal healing is crucial for ocular therapeutics.

    Purpose of the Study:

    • To investigate the impact of topical 0.5% Timoptic on corneal epithelial wound healing in rabbits.
    • To differentiate the effects of timolol maleate from the vehicle component.

    Main Methods:

    • Comparison of corneal reepithelialization rates in rabbits treated with Timoptic, vehicle, phosphate-buffered saline (PBS), PBS with timolol maleate, or no treatment.
    • Assessment of epithelial DNA content and cellular structure via light microscopy.
    • Measurement of DNA synthesis and cyclic adenosine monophosphate (cAMP) levels in epithelial cells.

    Main Results:

    • Both Timoptic and vehicle treatments slowed epithelial wound closure and reduced epithelial DNA content.
    • Light microscopy revealed fewer cellular layers and epithelial disruption in Timoptic- and vehicle-treated corneas.

    • The impaired corneal wound closure observed with Timoptic and vehicle was primarily attributed to benzalkonium chloride in the vehicle.
    • The results suggest that the vehicle, not the timolol maleate, is responsible for the observed delay in reepithelialization.
    • Further studies should consider the role of preservatives in topical ophthalmic formulations affecting corneal healing.
