Caffeine down-regulates beta adrenoreceptors in rat forebrain.

M R Goldberg, P W Curatolo, C S Tung

    Neuroscience Letters
    |July 20, 1982
    PubMed
    Summary

Caffeine treatment in female rats increased norepinephrine utilization in the brain. This was linked to a decrease in beta-adrenergic receptors, impacting neurotransmitter systems.

    Area of Science:

    • Neuropharmacology
    • Neurochemistry

    Background:

    • Caffeine is a widely consumed stimulant with known effects on the central nervous system.
    • Understanding caffeine's impact on neurotransmitter systems and receptor binding is crucial for its pharmacological characterization.

    Purpose of the Study:

    • To investigate the effects of caffeine on catecholamine utilization and adrenergic receptor binding in female rats.
    • To determine if caffeine alters norepinephrine and dopamine levels and adrenergic receptor density.

    Main Methods:

    • Female rats received caffeine (50 mg/kg) for three doses.
    • Catecholamine levels were measured after alpha-methyl-p-tyrosine administration.
    • Adrenergic receptor binding was assessed using radioligand assays ([3H]dihydroalprenolol, [3H]prazosin, [3H]clonidine).

    Main Results:

    • Caffeine enhanced the reduction in forebrain norepinephrine levels but not dopamine levels.
    • A significant reduction in beta-receptor density (Bmax) was observed.
    • No significant changes were found in alpha1 or alpha2 adrenergic receptor binding.

    Conclusions:

    • Caffeine selectively increases norepinephrine utilization in the rat forebrain.
    • This increased utilization is associated with a reduction in beta-adrenergic receptor density.
