Isotype switching in murine pre-B cell lines.

    Summary

Isotype switching occurs in pre-B cells via CH gene deletion, not requiring switch regions. This process can happen before V region formation, indicating prior mu chain expression isn

    Area of Science:

    • Immunology
    • Molecular Biology
    • Cell Biology

    Background:

    • Isotype switching is a critical process in B cell development.
    • The exact timing and mechanisms of isotype switching in early B cell stages are not fully understood.

    Purpose of the Study:

    • To investigate isotype switching at the pre-B cell stage.
    • To elucidate the molecular mechanisms involved in early B cell isotype commitment.

    Main Methods:

    • Utilized Abelson murine leukemia virus (A-MuLV)-transformed pre-B cell lines.
    • Established gamma 2b-producing clones from non-producing and mu-producing parent lines.
    • Performed Southern blot analysis to compare gene structures.

    Main Results:

    • Demonstrated that isotype switching can occur at the pre-B cell stage.
    • Identified a CH gene deletion mechanism for isotype switching, independent of switch regions.
    • Showed that deletion of intervening CH genes can precede functional V region formation.

    Conclusions:

    • Prior expression of mu chains is not mandatory for other isotype expression.
    • Isotype commitment can occur in pre-B cells.
    • A novel mechanism of isotype switching via CH gene deletion operates at the pre-B cell stage.
