[The lipocorticotropic system and prolactin secretion].

N Frisina, A Lasco, M Buemi

    Annales D'Endocrinologie
    |January 1, 1983

    Summary

Adrenocorticotropic hormone (ACTH) and dexamethasone reduce prolactin release by influencing the lipocorticotropic system and beta-endorphin. This suggests a feedback mechanism modulating prolactin secretion in healthy subjects.

    Area of Science:

    • Endocrinology
    • Neuroendocrinology
    • Pharmacology

    Background:

    • Prolactin secretion is regulated by complex neuroendocrine pathways.
    • The lipocorticotropic system and beta-endorphins are implicated in modulating hormone release.
    • Understanding these interactions is crucial for managing endocrine disorders.

    Purpose of the Study:

    • To investigate the role of the lipocorticotropic system in regulating prolactin secretion.
    • To determine the effect of ACTH and dexamethasone on prolactin response to stimulation.
    • To explore the involvement of beta-endorphins in prolactin modulation.

    Main Methods:

    • Administration of ACTH beta 1-24 (0.25 mg, i.m.) in healthy subjects.
    • Administration of dexamethasone (2 mg, orally) prior to insulin-induced hypoglycemia.
    • Administration of intravenous naloxone (0.4 mg/Kg) during insulin-induced hypoglycemia.

    Main Results:

    • ACTH administration reduced the prolactin response to sulpiride.
    • Dexamethasone blocked the prolactin response to insulin-induced hypoglycemia.
    • Naloxone blocked the prolactin response to insulin-induced hypoglycemia.

    Conclusions:

    • ACTH and dexamethasone likely influence the lipocorticotropic system via feedback.
    • Beta-endorphin release, modulated by the lipocorticotropic system, may regulate prolactin secretion.
    • Inhibition of the lipocorticotropic system may reduce prolactin system responsiveness to stimuli.
