    Summary

    Pinacidil tablets effectively lowered blood pressure in hypertensive patients over 9 weeks. The drug demonstrated a good safety profile with mild side effects, achieving target blood pressure for all participants.

    Background:

    • Hypertension is a prevalent cardiovascular condition requiring effective treatment.
    • Vasodilator drugs are a key therapeutic class for managing elevated blood pressure.

    Purpose of the Study:

    • To evaluate the efficacy and safety of a tablet formulation of pinacidil in patients with mild to moderate hypertension.
    • To assess the impact of pinacidil on blood pressure, pulse rate, and other physiological parameters.

    Main Methods:

    • A 9-week open-label study involving ten patients with mild to moderate hypertension.
    • Treatment with pinacidil tablets at daily dosages ranging from 40-100 mg, divided into two or three doses.
    • Monitoring of supine blood pressure, supine pulse rate, plasma urea, and body weight.

    Main Results:

    • Significant reduction in mean supine blood pressure from 174/100 to 148/84 mm Hg.
    • A modest increase in mean supine pulse rate of 3 beats/min.
    • Mild and frequent side effects were observed, typical of vasodilators; however, no patients withdrew due to adverse events.
