    Summary

Cell deformability, measured by filtrability, correlates with agglutinability by concanavalin A in mouse 3T3 and SV-3T3 cells. This finding predicts cell agglutination behavior based on cell filtration properties.

    Area of Science:

    • Cell Biology
    • Biophysics

    Background:

    • Previous studies suggested a link between cell deformability and agglutination.
    • Cell deformability is a critical factor in cell behavior and interactions.

    Purpose of the Study:

    • To investigate the correlation between cell deformability (filtrability) and agglutinability by concanavalin A.
    • To determine if cell filtrability can predict concanavalin A-induced agglutination in 3T3 and SV-3T3 cells.

    Main Methods:

    • Cell deformability was assessed using polycarbonate membrane filtration (filtrability).
    • Agglutinability was measured by the response of cells to concanavalin A.
    • Experiments involved EDTA-dissociated, trypsin-dissociated, and formalin-fixed 3T3 and SV-3T3 cells.

    Main Results:

    Related Experiment Videos

    • EDTA-dissociated cells showed low filtrability and low agglutinability, while trypsin-dissociated cells exhibited high filtrability and high agglutinability.
    • Formalin fixation drastically reduced both filtrability and agglutinability.
    • Admixing cells with different properties allowed for separation based on filtrability, confirming the correlation.

    Conclusions:

    • Cell filtrability is a reliable indicator of concanavalin A agglutinability in mouse 3T3 and SV-3T3 cells.
    • The study establishes a predictive relationship between cell mechanical properties and lectin-induced aggregation.
