    Background:

    • Prolactin secretion is a complex process regulated by various intracellular signaling pathways.
    • Calmodulin, a key calcium-binding protein, is implicated in numerous cellular functions, including hormone release.

    Purpose of the Study:

    • To investigate the potential involvement of calmodulin in the regulation of prolactin secretion.
    • To compare the inhibitory effects of various agents on prolactin release and calmodulin activity.

    Main Methods:

    • Utilized isolated rat anterior pituitary cells to assess prolactin secretion.
    • Employed an in-vitro system with calmodulin-activated cyclic GMP phosphodiesterase to measure calmodulin activity.
    • Compared the potencies of phenothiazines and N-(6-aminohexyl)-5-chloro-1-naphthalene sulfonamide (W7) in both systems.

    Main Results:

    • A strong correlation was observed between the inhibitory potencies of several phenothiazines and W7 on prolactin secretion and calmodulin activity.
    • Some drugs inhibited both prolactin secretion and calmodulin activity, supporting a role for calmodulin.
    • However, several drugs were more potent inhibitors of prolactin secretion than calmodulin, indicating involvement of other intracellular mechanisms.

    Conclusions:

    • Calmodulin is suggested to be involved in the control of prolactin secretion.
    • The findings indicate that while calmodulin plays a role, other intracellular systems also contribute to the regulation of prolactin release.
