    Summary

    A new Ig-Lp test detects immune complexes in autoimmune hyperlipidemia (AIH). This method is suitable for large-scale screening of AIH by identifying immunoglobulins (Ig) bound to low-density lipoproteins (Lp).

    Background:

    • Autoimmune hyperlipidemia (AIH) involves immunoglobulins (Ig) bound to lipoproteins (Lp) forming immune complexes.
    • Previous diagnostic methods for AIH are complex and not suitable for population screening.

    Purpose of the Study:

    • To develop and validate a novel, simpler Ig-Lp test for screening antilipoprotein autoimmune hyperlipidemia.
    • To assess the utility of the Ig-Lp test in identifying AIH in patient populations.

    Main Methods:

    • The Ig-Lp test detects immunoglobulins bound to low-density lipoproteins (LDL).
    • Method involves ultracentrifugation, delipidation, and protein quantification using radial immunodiffusion and laser immunonephelometry.
    • Antisera specific for IgG, IgA, IgM, LDL, and albumin were used.

    Main Results:

    • The Ig-Lp test identified 5 cases of hyperlipidemia associated with myeloma, previously classified as AIH.
    • The test showed specificity for the immunoglobulin type (IgA, IgG, IgM) involved in each case.
    • The test was negative in normolipidemic myelomas, healthy donors, and post-prandial normal serum.

    Conclusions:

    • The developed Ig-Lp test is a viable method for screening antilipoprotein autoimmune hyperlipidemia.
    • While not exclusively specific for antilipoprotein antibodies, the test's efficiency makes it suitable for large-scale AIH screening.
