Mutants of Corynebacterium diphtheriae PW8 that produce toxin in medium with excess iron
Applied and Environmental Microbiology
|December 1, 1981
Abstract:
Two mutants that produce toxin in medium with excess iron were isolated from the PW8 strain of Corynebacterium diphtheriae. These mutants produced as much toxin in medium containing excess iron (3 mug of Fe2+ per ml) as did the parent PW8 strain in iron-depleted medium, and they will be very useful for easy production of toxin.
Insights
Two Corynebacterium diphtheriae mutants were developed to produce toxin even with excess iron. These mutants simplify toxin production, making it more efficient for research and potential applications.
Area of Science:
- Microbiology
- Bacterial Genetics
- Toxin Production
Background:
- Corynebacterium diphtheriae is a bacterium that produces diphtheria toxin.
- Iron concentration significantly influences diphtheria toxin production.
- Optimizing toxin yield is crucial for research and therapeutic applications.
Purpose of the Study:
- To isolate and characterize mutants of Corynebacterium diphtheriae capable of producing toxin under iron-rich conditions.
- To assess the feasibility of using these mutants for simplified and efficient toxin production.
Main Methods:
- Isolation of mutants from the PW8 strain of Corynebacterium diphtheriae.
- Culturing mutants in media with varying iron concentrations.
- Quantification of toxin production in isolated mutants.
Main Results:
- Two mutants were successfully isolated that produce toxin in the presence of excess iron.
- These mutants exhibited toxin production levels comparable to the wild-type strain in iron-depleted conditions.
- The isolated mutants demonstrate robust toxin synthesis even when iron is abundant.
Conclusions:
- The developed mutants offer a valuable tool for the straightforward and cost-effective production of diphtheria toxin.
- These mutants overcome the iron-mediated repression of toxin synthesis, simplifying production protocols.
