Platelet size as a determinant of platelet function.

C B Thompson, J A Jakubowski, P G Quinn

The Journal of Laboratory and Clinical Medicine
    |February 1, 1983

    Summary

Platelet size, indicated by mean platelet volume (MPV), correlates with absolute platelet function, but not intrinsic function. Larger platelets release more granular content and aggregate more effectively.

    Area of Science:

    • Hematology
    • Platelet Physiology

    Background:

    • Mean platelet volume (MPV) is a measure of average platelet size.
    • Platelet subpopulations vary in size and may exhibit differential functions.

    Purpose of the Study:

    • To investigate the relationship between MPV and human platelet function.
    • To determine if platelet size influences aggregation, ATP, and beta-thromboglobulin (beta-TG) release.

    Main Methods:

    • Human platelets were separated into size-dependent subpopulations using counterflow centrifugation.
    • Platelet aggregation was induced by collagen and thrombin.
    • ATP and beta-TG content and release were measured in different platelet fractions.

    Main Results:

    • MPV showed a negative correlation with aggregation onset and positive correlations with aggregation rate and extent for collagen and thrombin.

    Related Experiment Videos

  • Ristocetin-induced agglutination was similar across all fractions.
  • Absolute release of ATP and beta-TG increased with MPV, but percent release remained constant.
  • Conclusions:

    • Intrinsic platelet function is similar across different sizes.
    • Absolute platelet function, including aggregation and granular content release, is positively correlated with platelet size (MPV).
