    Summary

    Background:

    • Reports to the Committee on Safety of Medicines (CSM) suggested mianserin might be more epileptogenic than tricyclic antidepressants.
    • This hypothesis was based on 40 reported cases of convulsions during mianserin treatment.

    Purpose of the Study:

    • To investigate the relative epileptogenic potential of mianserin compared to tricyclic antidepressants.
    • To assess the reliability of CSM data for evaluating drug-induced seizures.

    Main Methods:

    • A questionnaire study collected data on 83% of reported mianserin-related convulsion cases.
    • Case data were compared with a control group.
    • Neurologist and expert ratings assessed the drug-effect relationship, revealing variability.

    • CSM data alone were insufficient for a reliable assessment of relative epileptogenic effects.
