    Background:

    • Platelet size is a variable trait with implications for hemostasis and thrombotic disorders.
    • Previous studies suggest a correlation between platelet size and age, but the underlying mechanisms remain unclear.

    Purpose of the Study:

    • To investigate the relationship between platelet size and in vivo aging in baboons.
    • To determine if platelet size heterogeneity is established at production or during circulation.

    Main Methods:

    • Separation of baboon platelets into size-dependent subpopulations using counterflow centrifugation.
    • In vivo and in vitro labeling of platelets with 75Se-methionine, 51Cr, and 14C-serotonin.
    • Analysis of platelet radioactivity, mean platelet volume (MPV), lactate dehydrogenase (LDH) activity, and dense-body content.

    Main Results:

    • Platelet subpopulations exhibited characteristics similar to human platelets.
    • A strong correlation was observed between MPV and platelet radioactivity across all labeling methods.
    • Platelet survival studies indicated that larger platelets have a longer lifespan, with no evidence of label translocation between fractions.

    • Platelet size heterogeneity is established at the time of release from the bone marrow due to production factors.
    • Larger platelets are associated with a longer lifespan, not necessarily younger age.
    • These findings challenge the notion of platelet maturation in circulation as a determinant of size.
