    Summary

    Primary hyperparathyroidism may alter immune responses. This study found that plasma from patients with this condition enhances lymphocyte proliferation, suggesting a humoral factor influences immune cell activity.

    Background:

    • Primary hyperparathyroidism is linked to increased malignancy risk, suggesting altered immune function.
    • The specific nature of immune dysregulation in primary hyperparathyroidism remains unclear.

    Purpose of the Study:

    • To investigate lymphocyte proliferation responses to mitogens in primary hyperparathyroidism.
    • To determine if plasma from patients with primary hyperparathyroidism affects lymphocyte activation.

    Main Methods:

    • Isolated lymphocytes from 6 primary hyperparathyroidism patients and 4 controls.
    • Stimulated lymphocytes with phytohemagglutinin (PHA), concanavalin A (Con-A), and pokeweed mitogen (PWM).
    • Assessed lymphocyte proliferation via [3H]thymidine incorporation in the presence of autologous or normal AB+ plasma.

    Main Results:

    • Lymphocyte proliferation was significantly enhanced when stimulated with mitogens in the presence of plasma from primary hyperparathyroidism patients, irrespective of lymphocyte source.
    • Both patient and normal lymphocytes showed increased proliferation with patient plasma.

    Conclusions:

    • Plasma from primary hyperparathyroidism patients contains a factor that potentiates lymphocyte mitogenesis.
    • Alternatively, normal plasma may contain an inhibitor of lymphocyte activation that is absent in patient plasma.
    • These findings indicate a humoral influence on immune cell activity in primary hyperparathyroidism.
