    Background:

    • CBA/N mice exhibit an X-linked immunodeficiency.
    • This deficiency is characterized by impaired humoral immune responses, particularly to sheep red blood cells (SRBC).

    Purpose of the Study:

    • To investigate the cellular mechanisms underlying the deficient humoral response in CBA/N mice.
    • To specifically examine helper T cell function in an adoptive transfer system.

    Main Methods:

    • Utilized an adoptive transfer system with different sources of helper T cells.
    • Compared helper T cell function using thymocytes versus radioresistant helper T cells from primed, irradiated recipients.
    • Assessed antibody production (plaque-forming cells, PFC) against SRBC.

    • CBA/N thymocytes were effective precursors for helper T cells.
    • However, radioresistant helper T cells from primed, irradiated CBA/N mice showed significantly reduced function compared to CBA/Ca controls.

    • The observed deficiency is not attributed to suppressor T cell activity or negative environmental effects on B cells or T-B cell cooperation.
    • The radiosensitivity of helper T cells is a key feature of this X-linked immunodeficiency.
