    Summary

    High glucose levels do not impair polymorphonuclear leukocyte (PMNL) function in vitro. These findings suggest that elevated blood sugar alone is not the cause of PMNL dysfunction in diabetes mellitus patients.

    Background:

    • Polymorphonuclear leukocytes (PMNLs) are critical immune cells.
    • PMNL dysfunction is observed in patients with diabetes mellitus.
    • The role of hyperglycemia in PMNL dysfunction requires clarification.

    Purpose of the Study:

    • To investigate the impact of varying glucose concentrations on PMNL oxidative metabolism and functional activities.
    • To determine if hyperglycemic conditions affect PMNL oxygen consumption, superoxide generation, phagocytosis, and bacterial killing.

    Main Methods:

    • PMNLs were exposed to glucose concentrations from 100-600 mg/dl for different incubation periods.
    • Assays were performed to measure oxygen consumption and superoxide generation.
    • Phagocytosis and killing of radiolabeled Staphylococcus aureus by PMNLs were assessed.

    Main Results:

    • PMNL oxidative metabolism and functional activities remained unaffected by hyperglycemic conditions.
    • No significant changes were observed in oxygen consumption, superoxide generation, phagocytosis, or bacterial killing.
    • These effects were consistent for incubation periods up to 18 hours.

    Conclusions:

    • Elevated glucose concentrations in vitro do not impair PMNL oxidative metabolism or functional capacity.
    • Hyperglycemia per se does not appear to be the direct cause of PMNL abnormalities seen in diabetic patients.
    • Further research is needed to identify the specific factors contributing to PMNL dysfunction in diabetes mellitus.
