    Background:

    • Dipyridamole, a nucleoside transport inhibitor, was previously reported to stimulate endothelial cell proliferation in rats.
    • The direct angiogenic potential of dipyridamole requires further investigation.

    Purpose of the Study:

    • To investigate the direct effects of dipyridamole on endothelial cell proliferation and migration in vitro.
    • To evaluate the angiogenic potential of dipyridamole using the chorioallantoic membrane (CAM) model.

    Main Methods:

    • Cultured calf aortic endothelial cells were used to assess proliferation and migration.
    • The chorioallantoic membrane (CAM) model was employed to observe vascular responses to dipyridamole and its pharmaceutical preparation, Curantyl.
    • pH levels were manipulated to determine their influence on observed vascular effects.

    Main Results:

    • Dipyridamole did not directly stimulate the proliferation or migration of cultured calf aortic endothelial cells.
    • The pharmaceutical preparation Curantyl induced vascular responses in the CAM model.
    • These vascular responses were dependent on the low pH of the Curantyl preparation and could be abolished by increasing the pH.

    Conclusions:

    • Dipyridamole does not appear to directly stimulate endothelial cell proliferation or migration.
    • The observed vascular effects of Curantyl in the CAM model are likely due to its acidic nature, not a direct angiogenic effect of dipyridamole.
    • Further research is needed to clarify the in vivo angiogenic effects of dipyridamole.
