    Background:

    • Chlorpropamide, a sulfonylurea drug, has been anecdotally suggested to possess antidiuretic properties.
    • The mechanism by which chlorpropamide might influence water balance, specifically antidiuretic hormone (ADH) release, requires investigation.

    Purpose of the Study:

    • To investigate the effect of chlorpropamide on the release of antidiuretic hormone (ADH) in healthy human volunteers.
    • To quantify arginine vasopressin (AVP) levels following intravenous chlorpropamide administration.

    Main Methods:

    • Intravenous administration of chlorpropamide to nine healthy, fasted volunteers.
    • Comparison of ADH/AVP levels with a placebo (saline) control.
    • Measurement of serum chlorpropamide concentrations to determine pharmacokinetic profile.

    Main Results:

    • No evidence of increased ADH release was observed after chlorpropamide administration compared to placebo.
    • Arginine vasopressin (AVP) levels were slightly lower in the first 2 hours post-chlorpropamide injection than after saline.
    • Pharmacokinetic analysis revealed a rapid distribution phase followed by a slow elimination phase, with a distribution half-life (t 1/2) of 1.00 ± 0.26 hours.

    Conclusions:

    • Chlorpropamide does not stimulate ADH release.
    • The observed slight decrease in AVP suggests chlorpropamide may not have a significant antidiuretic effect via ADH stimulation.
    • The pharmacokinetic profile of chlorpropamide was characterized in healthy volunteers.
