    Background:

    • Thiamine (Vitamin B1) is essential for carbohydrate metabolism and cellular energy production.
    • Furosemide is a potent loop diuretic commonly prescribed for edema and hypertension.
    • Potential interactions between diuretics and vitamin levels are not fully understood.

    Purpose of the Study:

    • To investigate the effect of long-term furosemide administration on thiamine levels in rats.
    • To determine if furosemide influences thiamine excretion and utilization.
    • To assess the impact of furosemide on transketolase activity, an indicator of thiamine status.

    Main Methods:

    • Male Wistar rats were divided into four groups: control (thiamine-sufficient), furosemide-treated (thiamine-sufficient), thiamine-deficient, and furosemide-treated (thiamine-deficient).
    • Rats received daily intraperitoneal injections of furosemide (20 mg/kg) or saline for four weeks.
    • Thiamine concentrations, transketolase activity, and thiamine pyrophosphate effect were measured in blood and various tissues.
    • Urinary thiamine excretion was assessed after single administrations of different furosemide doses (20, 10, and 2 mg/kg).

    • Furosemide administration significantly decreased thiamine concentration and transketolase activity in both thiamine-sufficient and deficient rats compared to controls.
