    Background:

    • Prostaglandins (PGs) are lipid compounds with diverse physiological roles.
    • The role of PGs in cardiac automaticity, particularly in mammalian pacemaker tissue, requires further elucidation.

    Purpose of the Study:

    • To investigate the direct effects of prostaglandins on sinoatrial (SA) nodal automaticity in guinea pig heart preparations.
    • To explore the potential endogenous prostaglandin system modulating heart rate.

    Main Methods:

    • Isolated guinea pig SA-nodal and right atrial preparations were utilized.
    • Prostaglandins (PGE2, PGE1, PGF2alpha) were administered in nanomolar concentrations.
    • Indomethacin (prostaglandin synthesis inhibitor) and propranolol (beta-adrenergic blocker) were used for pretreatment.

    Main Results:

    • Prostaglandins PGE2, PGE1, and PGF2alpha significantly increased SA nodal automaticity in a dose-dependent manner.
    • Pretreatment with indomethacin reduced spontaneous heart rate, suggesting an endogenous PG system.
    • Guinea pig tissue showed direct positive chronotropic responses to PGE2, independent of beta-adrenergic receptors.

    Conclusions:

    • Exogenous prostaglandins, particularly PGE2, directly stimulate guinea pig cardiac pacemaker tissue at very low doses.
    • An endogenous, sensitive prostaglandin system appears to play a crucial role in modulating heart rate in guinea pigs.
    • Guinea pig SA-nodal tissue exhibits a distinct response to PGE2 compared to rabbit tissue.
