    Background:

    • B-lymphocytes express receptors for the third component of complement (C3).
    • Rosette formation with C3-coated erythrocytes identifies B-lymphocytes.

    Purpose of the Study:

    • To investigate the binding characteristics of C3 and its fragment C3b to human tonsil lymphocytes.
    • To determine if C3 is cleaved to C3b upon binding to lymphocytes.

    Main Methods:

    • Rosette formation assays using sheep erythrocytes coated with antibody and complement components (EAC143b).
    • Inhibition assays with fluid-phase C3 and C3b.
    • Binding studies using radioiodinated C3 and C3b.
    • Inhibition of binding using salicylhydroxamic acid.

    Main Results:

    • Fluid-phase C3 and C3b similarly inhibited rosette formation.
    • C3 was not cleaved to C3b during incubation with lymphocytes.
    • Both radioiodinated C3 and C3b exhibited specific binding to lymphocytes.
    • Salicylhydroxamic acid did not inhibit C3 binding, unlike C3b binding.

    Conclusions:

    • Human tonsil B-lymphocytes bind both C3 and C3b.
    • C3 and C3b share a common binding characteristic for lymphocyte receptors.
    • C3 binding to lymphocytes does not involve cleavage to C3b.
