Background:

  • Epithelium-lining macrophages are spindle-shaped cells found in the epidermis and hair follicles.
  • Their role in skin conditions like psoriasis has been largely unstudied.

Purpose of the Study:

  • To investigate the distribution and immunophenotype of epithelium-lining macrophages in normal and psoriatic skin.
  • To determine if these macrophages represent a distinct subset of dermal monocytes/macrophages.

Main Methods:

  • Analysis of 25 lesional psoriatic and 5 normal skin biopsies.
  • Immunophenotyping using markers such as CD45, CD14, CD36, CD4, CD11a, HLA-DR, CD1, CD34, and factor XIIIa.

Main Results:

  • Epithelium-lining macrophages were inconspicuous in normal skin but increased in psoriatic lesions.
  • These cells formed a continuous row at the dermo-epidermal junction in some psoriatic biopsies.
  • Immunophenotyping revealed expression of CD45, CD14, CD36, CD4, HLA-DR, and CD11a, but not CD11b, CD1, or CD34.

Conclusions:

  • Epithelium-lining macrophages are a distinct subset of dermal monocytes/macrophages with unique localization and immunophenotype.
  • Their association with the epidermis suggests a role in regulating epidermal growth.
  • Expression of immune molecules indicates potential involvement in T cell activation.
