Low antibody binding capacity (ABC) CD20 expression was found on T-acute lymphoblastic leukemia (T-ALL) cells. This finding helps differentiate T-cell malignancies from normal T cells and aids in monitoring disease recurrence.

Area of Science:

  • Immunology
  • Hematology
  • Oncology

Background:

  • CD20 is typically a B lineage restricted antigen.
  • Recent studies have detected low antibody binding capacity (ABC) CD20 on a subset of normal T lymphocytes.

Observation:

  • This study reports low ABC CD20 expression in three cases of T-acute lymphoblastic leukemia (T-ALL).
  • Immunophenotypic analysis revealed a pure population of malignant T cells expressing CD7 and low ABC CD20.
  • Other B lymphocyte associated antigens were negative.

Findings:

  • Three cases of T-ALL exhibited low ABC CD20 expression (95%, 87%, 79% of malignant cells).
  • This contrasts with the bright CD20 expression typically seen on normal B lymphocytes.
  • Eleven other T-lymphocytic malignancies tested were CD20 negative.

Related Experiment Videos

Implications:

  • Low ABC CD20 expression can serve as a unique immunophenotypic marker for T-cell neoplasia.
  • This marker aids in distinguishing T-cell malignancies from normal or reactive T cells.
  • The presence of low ABC CD20 may facilitate monitoring for residual or recurrent disease in T-ALL patients.
