Summary

Pregnancy naturally immunizes T cells against breast cancer mucin (MUC-1), potentially protecting against cancer. This suggests MUC-1 peptides could be used in breast cancer vaccines.

Background:

  • Multiparity (having multiple children) is associated with a reduced risk of breast cancer.
  • The human breast cancer-associated mucin (MUC-1) is a known tumor marker and potential therapeutic target.

Purpose of the Study:

  • To investigate the immunological response to MUC-1 during pregnancy.
  • To explore the potential of MUC-1 as a target for breast cancer prevention and therapy.

Main Methods:

  • T cell proliferation assays were performed using peripheral blood mononuclear cells (PBMCs) from multiparous women, nulliparous women, and men.
  • T cells were stimulated with core peptide sequences of MUC-1.
  • PBMCs from two nulliparous women were retested during their first trimester of pregnancy.

Main Results:

  • T cells from multiparous women demonstrated specific proliferation in response to MUC-1 peptide sequences.
  • T cells from nulliparous women and men did not show this specific proliferation.
  • T cells from nulliparous women who became pregnant showed specific MUC-1 reactivity during early pregnancy.

Conclusions:

  • Pregnancy may induce a natural immunization against MUC-1 epitopes, contributing to breast cancer protection.
  • Synthetic MUC-1 peptides show promise as components for breast cancer vaccines for both therapeutic and preventive strategies.
