A new hepatocyte stimulating factor: cardiotrophin-1 (CT-1)
11st Department of Medicine, Johannes Gutenberg University of Mainz, Germany.
Cardiotrophin-1 (CT-1), a novel cytokine, stimulates liver cells to produce acute phase proteins. This finding identifies CT-1 as a significant hepatocyte stimulating factor within the IL-6 cytokine family.
Area of Science:
- Biochemistry
- Molecular Biology
- Cell Biology
Background:
- Cardiotrophin-1 (CT-1) is a newly identified cytokine.
- CT-1 shares sequence similarity with the IL-6/LIF/CNTF/OSM/IL-11 cytokine family.
- Members of the IL-6 cytokine family are known to induce hepatic acute phase protein (APP) gene expression.
Purpose of the Study:
- To investigate the potential of CT-1 to induce a liver acute phase response.
- To determine if CT-1 functions as a hepatocyte stimulating factor.
Main Methods:
- Stimulation of rat hepatoma cells with CT-1 and other cytokines (LIF, OSM, IL-6).
- Measurement of rat fibrinogen mRNA expression.
- Stimulation of human hepatoma cells and primary rat hepatocytes with CT-1.
- Analysis of human haptoglobin and rat alpha 2-macroglobulin mRNA expression.
Main Results:
- CT-1 and LIF strongly induced rat fibrinogen mRNA expression in rat hepatoma cells.
- OSM and IL-6 showed a less pronounced induction of fibrinogen mRNA.
- CT-1 induced human haptoglobin and rat alpha 2-macroglobulin mRNA expression in human and primary rat cells, respectively.
- The induction of the acute phase response by CT-1 was dose- and time-dependent.
Conclusions:
- CT-1 is a potent inducer of the hepatic acute phase response.
- CT-1 acts as a hepatocyte stimulating factor.
- CT-1's function aligns with its classification within the IL-6 cytokine family.
