Background:

  • Central nervous system (CNS) depressant drugs are known to affect various physiological processes.
  • Altered feeding behavior, including hyperphagia, can be a side effect of certain CNS medications.

Purpose of the Study:

  • To investigate the role of peptidergic mechanisms in drug-induced hyperphagia in mice.
  • To determine if naloxone can reverse hyperphagia caused by CNS depressants.

Main Methods:

  • Hyperphagia was induced in mice using oral administration of CNS depressant drugs: chlordiazepoxide, diazepam, cyproheptadine, and phenobarbitone.
  • Mice were pretreated with naloxone (subcutaneously) to assess its effect on drug-induced hyperphagia.
  • Food intake was measured to quantify the effects of the drugs and naloxone.

Main Results:

  • Oral administration of chlordiazepoxide, diazepam, cyproheptadine, and phenobarbitone successfully induced hyperphagia in mice.
  • Pretreatment with naloxone (0.1 mg/kg) abolished the hyperphagic effect of these CNS depressant drugs.

  • Naloxone's ability to reverse drug-induced hyperphagia indicates a potential interaction between opioid and peptidergic systems in appetite control.
