Purpose of the Study:

  • To isolate and characterize the cDNA encoding a 20 kD epididymal protein (EP20).
  • To investigate the potential role of EP20 in sperm function.

  • Polyclonal antibodies against EP20 were used to screen a human testis lambda gt11 expression library.
  • Nucleotide sequencing of the isolated cDNA was performed.
  • Sequence homology analysis was conducted against existing databases.

  • A cDNA sequence of 1908 bp was isolated, containing an open reading frame encoding a 493-amino acid polypeptide.
  • The EP20 cDNA sequence showed high identity (97%) to a human EST clone (HHC M14) in its 3' untranslated region.
  • The deduced amino acid sequence of EP20 showed no homology to known polypeptides.

Conclusions:

  • A novel human epididymal protein, EP20, has been identified and characterized at the cDNA level.
  • EP20 may play a role in sperm maturation and/or capacitation.
