Insights

The DEAD box protein DDX1 gene is frequently co-amplified with the MYCN gene in neuroblastoma. This co-amplification may indicate a poor prognosis in patients with this cancer.

Area of Science:

  • Molecular Biology
  • Cancer Genetics

Background:

  • DEAD box proteins are RNA helicases involved in RNA processing.
  • The DDX1 gene, containing a DEAD box motif, is located at chromosome band 2p24.
  • MYCN amplification is a known indicator of poor prognosis in neuroblastoma.

Observation:

  • The DDX1 gene is co-amplified with the MYCN gene in retinoblastoma cell lines.
  • DDX1 gene co-amplification with MYCN was observed in 2 of 3 neuroblastoma cell lines.
  • DDX1 RNA levels correlated with DDX1 gene copy number in neuroblastoma.

Findings:

  • Of 32 neuroblastoma samples, 13 had MYCN amplification.
  • Seven of the 13 MYCN-amplified neuroblastomas also showed DDX1 amplification.
  • No DDX1 amplification was observed in neuroblastomas without MYCN amplification.

Implications:

  • This is the first reported instance of frequent co-amplification between DDX1 and MYCN in neuroblastoma.
  • Co-amplification of DDX1 and MYCN may have prognostic significance in neuroblastoma.
  • Further studies are needed to confirm the prognostic value of DDX1 co-amplification due to small sample size.
