High-performance liquid chromatography, or HPLC, is an analytical technique that separates liquid samples under high pressures. An HPLC instrument consists of glass bottles for storing solvents called mobile phase reservoirs. HPLC-grade solvents are used to maintain high purity, and the dissolved gases are removed using a degasser, such as a vacuum pumping system or sparging with helium. The solvents are then pumped into the analytical column using a screw-driven syringe or reciprocating pumps.

Summary

Lithostathine concentration is decreased in patients with chronic calcifying pancreatitis (CCP). This protein, normally found in pancreatic juice, may be degraded by trypsin, affecting its measurement in CCP patients.

Background:

Purpose of the Study:

  • To investigate the discrepancies in lithostathine concentration measurements in CCP patients.
  • To develop a novel assay for accurate lithostathine measurement in pancreatic juice.
  • To determine the association between lithostathine levels and CCP.

Main Methods:

  • Developed a novel lithostathine assay using high-performance liquid chromatography (HPLC).
  • Separated secretory proteins based on hydrophobic interactions using a Phenyl-TSK column at pH 5.0.
  • Quantified lithostathine concentrations relative to total protein in pancreatic juice.

Main Results:

  • Conflicting immunoassay results may stem from lithostathine's susceptibility to trypsin cleavage and subsequent epitope modification.
  • Lithostathine concentrations (μg/mg total protein) were significantly lower in CCP patients (alcoholic: 6.3±2.7; other: 7.2±3.7) compared to controls (16.7±4.3).
  • Similar lithostathine concentrations were observed in chronic alcoholic patients without CCP (6.6±3.3) and CCP patients.

Conclusions:

  • Decreased lithostathine concentration is associated with chronic calcifying pancreatitis (CCP).
  • Lithostathine levels are reduced in CCP regardless of alcoholic etiology.
  • Reduced lithostathine concentration is linked to CCP but is not the sole cause of the disease.
