Summary

Interleukin-4 (IL-4) plays a key role in the proliferative aspects of intestinal immunopathology during graft-versus-host reactions. Targeting IL-4 may offer a novel therapeutic strategy for enteropathies.

Purpose of the Study:

  • To investigate the specific role of Interleukin-4 (IL-4) in the intestinal manifestations of graft-versus-host reactions (GVHR).
  • To determine if targeting IL-4 could prevent or mitigate intestinal immunopathology.

Main Methods:

  • Depletion of IL-4 using monoclonal antibodies (MoAb) or a soluble IL-4 receptor (IL-4R).
  • Assessment of jejunal pathology, including crypt hyperplasia and intraepithelial lymphocyte (IEL) recruitment, in a proliferative GVHR model.
  • Evaluation of villus atrophy in a destructive GVHR model and systemic immunity indices.

Main Results:

  • IL-4 depletion prevented jejunal manifestations (crypt hyperplasia, IEL recruitment) in a proliferative GVHR model.
  • IL-4 depletion did not affect villus atrophy in a destructive GVHR model.
  • Systemic immunity indices remained unaffected by IL-4 depletion.

Conclusions:

  • IL-4 selectively mediates proliferative aspects of intestinal immunopathology in GVHR.
  • IL-4 represents a potential therapeutic target for specific enteropathies characterized by proliferative changes.
