    Area of Science:

    • Neuroendocrinology
    • Psychiatry
    • Pharmacology

    Background:

    • The pineal neurohormone melatonin is investigated for its potential therapeutic effects.
    • Depression and Huntington's chorea are complex neurological and psychiatric conditions.

    Purpose of the Study:

    • To evaluate the efficacy of melatonin in treating moderate to severe depression.
    • To observe the effects of melatonin administration in patients with Huntington's chorea.

    Main Methods:

    • A double-blind crossover study design was employed.
    • Varying doses of melatonin were administered to eight patients (six with depression, two with Huntington's chorea).

    Main Results:

    • Melatonin administration exacerbated dysphoric symptoms in patients.
    • Observed side effects included sleep disturbance, weight loss, and decreased oral temperature.
    • Increased cerebrospinal fluid 5-hydroxyindoleacetic acid and calcium levels were noted in a subset of patients.

    Conclusions:

    • Melatonin may have adverse effects on mood and physiological functions in certain patient populations.
    • The observed biochemical changes warrant further investigation into melatonin's mechanism of action in neuropsychiatric disorders.
