Background:

  • Behçet's disease (BD) is characterized by activated polymorphonuclear leukocytes (PMNs) and their infiltration into lesions.
  • Interleukin 8 (IL-8) is a key cytokine known to activate PMNs.

Purpose of the Study:

  • To investigate the role of IL-8 in the pathogenesis of Behçet's disease.

Main Methods:

  • Examined IL-8 production in vitro and in vivo in 25 BD patients.
  • Utilized ELISA for IL-8 levels, Northern blot for mRNA expression, and polarization assays for PMN activity.
  • Immunohistochemistry was employed to detect IL-8 in BD lesions.

Main Results:

  • Elevated IL-8 levels and mRNA expression in peripheral mononuclear cells (MNCs) of active BD patients compared to controls.
  • PMNs from active BD patients showed accelerated activity but reduced response to exogenous IL-8.
  • IL-8 was detected in MNCs, endothelial cells, and fibroblasts within BD lesions.

Conclusions:

  • IL-8 production may be accelerated in Behçet's disease, potentially contributing to disease pathogenesis.
  • IL-8 plays a significant role in the inflammatory processes observed in BD lesions.
