Summary

This study investigated if clozapine causes tardive dyskinesia. While not definitive, clozapine appears to carry a lower risk of tardive dyskinesia compared to conventional neuroleptics.

Area of Science:

  • Psychiatry
  • Pharmacology

Background:

  • Investigating the potential link between clozapine and tardive dyskinesia (TD).
  • Assessing TD risk in patients with schizophrenia or schizoaffective disorder.

Purpose of the Study:

  • To determine if chronic clozapine exposure causes tardive dyskinesia.
  • To compare the incidence of TD in clozapine-treated patients versus those on conventional neuroleptics.

Main Methods:

  • Analysis of modified Simpson Dyskinesia Scale ratings in 28 patients treated with clozapine for at least 1 year.
  • Comparison with a control group of similarly diagnosed patients treated with conventional neuroleptics for at least 1 year.

Main Results:

  • Two patients on clozapine developed mild tardive dyskinesia, though causality was uncertain.

Related Experiment Videos

  • Survival analysis indicated a lower risk of developing tardive dyskinesia in the clozapine group compared to the neuroleptic group.
  • Conclusions:

    • The study could not definitively establish clozapine as a cause of tardive dyskinesia.
    • The risk of tardive dyskinesia appears lower with clozapine than with typical neuroleptics.
