    Summary

    Thyrotropin releasing hormone (TRH) showed a minimal, short-lived beneficial effect in depressed patients, but was not therapeutically valuable. TRH infusion did not alter thyroid hormone levels or TSH response in patients compared to controls.

    Background:

    • Thyrotropin releasing hormone (TRH) plays a role in regulating the pituitary-thyroid axis.
    • Depression is a complex mood disorder with potential neuroendocrine dysregulation.

    Purpose of the Study:

    • To investigate the therapeutic potential of a continuous thyrotropin releasing hormone (TRH) infusion in patients with depression.
    • To assess the impact of TRH infusion on thyroid-stimulating hormone (TSH) and thyroid hormone levels in depressed patients and healthy controls.

    Main Methods:

    • A double-blind, cross-over trial was conducted with 10 depressive patients and a control group.
    • Patients received a slow, continuous infusion of 1000 mug TRH over 4 hours or a placebo.

  • A faint, short-lasting beneficial effect of TRH infusion was observed in depressive patients, deemed of no therapeutic value.
  • No significant differences were found in TSH response, T3 resin uptake, T4, or free thyroxine index between depressive patients and controls following TRH infusion.

    • Continuous TRH infusion at the tested dosage and duration lacks therapeutic efficacy for depression.
    • TRH infusion does not appear to significantly alter thyroid function markers or TSH response in depressed individuals compared to normal subjects.
