Summary

N-glucuronidation of N-acetylbenzidine was demonstrated, forming an acid-labile N-acetylbenzidine-N’-glucuronide. This pathway is significant for N-acetylbenzidine metabolism, particularly in humans, aiding in detoxification.

Area of Science:

  • Pharmacokinetics and Metabolism
  • Toxicology
  • Carcinogenesis

Background:

  • N-glucuronidation is a key metabolic pathway for aromatic amines.
  • The N-glucuronidation of N-acetylbenzidine was previously uncharacterized.

Purpose of the Study:

  • To synthesize and identify N-acetylbenzidine-N'-glucuronide.
  • To investigate the formation of N-acetylbenzidine-N'-glucuronide in liver preparations from different species.
  • To compare N-glucuronidation rates of N-acetylbenzidine and benzidine.

Main Methods:

  • Synthesis and mass spectrometry identification of N-acetylbenzidine-N'-glucuronide.
  • Incubation of [3H]N-acetylbenzidine with liver slices and microsomes from human, dog, and rat.
  • Assessment of N-acetylbenzidine-N'-glucuronide and benzidine-N-glucuronide formation.

Related Experiment Videos

  • Evaluation of acid lability of the synthesized glucuronides.
  • Main Results:

    • N-acetylbenzidine-N'-glucuronide was synthesized and confirmed by mass spectrometry.
    • Significant formation of N-acetylbenzidine-N'-glucuronide occurred in human liver slices and microsomes.
    • N'-glucuronide formation was not detected in dog liver preparations but was observed in rat.
    • Relative glucuronidation activity followed the order: human >> dog > rat for both N-acetylbenzidine and benzidine.
    • N,N'-diacetylbenzidine did not undergo N-glucuronidation.

    Conclusions:

    • N-acetylbenzidine-N'-glucuronide formation is a significant metabolic pathway for N-acetylbenzidine, especially in humans.
    • The acid lability of these N-glucuronides suggests a role in hepatic detoxification and potential bladder accumulation of carcinogens.
    • N-acetylation influences the N-glucuronidation of aryldiamines, impacting bladder carcinogenesis.
