Summary

Investigating yeast autonomously replicating sequence ARS305 revealed critical DNA elements essential for chromosomal replication origin function. These elements, located downstream of the origin recognition complex binding site, are vital for efficient DNA replication.

Area of Science:

  • Molecular Biology
  • Yeast Genetics
  • DNA Replication

Background:

  • Autonomously replicating sequences (ARS) are crucial for DNA replication initiation in yeast.
  • ARS305 shares components with the chromosomal replicator ORI305, including an ARS consensus sequence (ACS) and a DNA unwinding element.

Purpose of the Study:

  • To identify and characterize functional DNA elements within ARS305 located 3' to the ACS.
  • To determine the role of these elements in both plasmid ARS function and chromosomal origin activity.

Main Methods:

  • Linker substitution mutagenesis of ARS305.
  • Plasmid mitotic stability assays.
  • Transplacement of mutations into the chromosomal ORI305 locus.
  • Two-dimensional gel electrophoresis to analyze replication intermediates.

Related Experiment Videos

Main Results:

  • Three functional sequence elements were identified downstream of the ACS in ARS305.
  • Mutation of the major element abolished chromosomal origin function.
  • Mutations in the other two elements reduced both plasmid and chromosomal replication activities.

Conclusions:

  • Multiple DNA elements downstream of the ACS are essential for ARS305 function in its chromosomal context.
  • While ORC-binding elements are conserved, the arrangement and extent of downstream elements vary among yeast replicators.
