    Background:

    • Hemoglobin alpha-chain heterogeneity was investigated in sika deer (Cervus nippon).
    • This research was prompted by observed alpha-chain gene locus heterogeneity in Virginia white-tailed deer (Odocoileus virginianus).

    Purpose of the Study:

    • To investigate the molecular basis of hemoglobin alpha-chain variations in sika deer.
    • To understand the evolutionary implications of observed alpha-chain diversity in Cervus nippon.

    Main Methods:

    • Electrophoresis was used to analyze hemoglobin chains from ten sika deer specimens.
    • Peptide mapping was employed to identify variations within specific alpha-chain peptides (alpha-TPIII and alpha-TPIV).

    Main Results:

    • Electrophoresis revealed a single alpha-chain band, but peptide mapping identified variations at positions 15, 20, and 22.
    • At least five distinct alpha-chains were identified, with substitutions including glycine, aspartic acid (Asp), asparagine/aspartic acid (Asx), lysine (Lys), proline (Pro), and serine.
    • The most common alpha-chain contained Asp-15, Lys-20, and Pro-22.

    Conclusions:

    • The extensive alpha-chain variability in sika deer is likely due to gene duplication events, possibly from interbreeding between subspecies.
    • Sika deer may have evolved from a lineage related to white-tailed deer after alpha-chain gene duplication in the latter.
    • These findings highlight the remarkable variability within the hemoglobin alpha-chain region in sika deer.
