Summary

Endoglin glycoprotein modulates transforming growth factor-beta (TGF-beta) signaling in monocytes. Overexpressing endoglin isoforms blocks TGF-beta 1-induced proliferation and c-myc downregulation, revealing its role in TGF-beta pathway regulation.

Area of Science:

  • Molecular Biology
  • Cell Biology
  • Immunology

Background:

  • Endoglin is a membrane glycoprotein binding transforming growth factor-beta (TGF-beta) isoforms.
  • Endoglin expression is upregulated during monocyte differentiation.
  • TGF-beta can stimulate endoglin expression in human monocytes and U-937 cells.

Purpose of the Study:

  • To investigate the functional role of endoglin in modulating cellular responses to TGF-beta.
  • To compare the effects of L- and S-endoglin isoforms on TGF-beta signaling.
  • To elucidate the mechanism of endoglin's action in TGF-beta pathways.

Main Methods:

  • Generation of U-937 cell transfectants overexpressing L- or S-endoglin isoforms.
  • Analysis of TGF-beta 1 and TGF-beta 2 effects on cell proliferation, c-myc mRNA, fibronectin synthesis, adhesion, PECAM-1 phosphorylation, and aggregation.

Related Experiment Videos

  • Assessment of ligand binding and degradation rates in endoglin transfectants.
  • Main Results:

    • Overexpression of L-endoglin abrogated TGF-beta 1-induced inhibition of proliferation and c-myc downregulation.
    • S-endoglin partially reduced these TGF-beta 1 effects.
    • Endoglin isoforms differentially affected TGF-beta 1 responses, including fibronectin synthesis and cell adhesion, without altering ligand binding or degradation significantly.

    Conclusions:

    • Endoglin plays a crucial role in mediating TGF-beta 1 signaling in monocytes.
    • Different endoglin isoforms exhibit distinct modulatory effects on TGF-beta responses.
    • Endoglin influences TGF-beta 1 cellular effects through mechanisms independent of ligand binding or degradation.
