Thymic epithelial cells (TECs) present antigens to T cells, crucial for T cell selection. Cortical TECs (cTECs) and medullary TECs (mTECs) show distinct antigen processing pathways, influencing T cell repertoire development.

Background:

  • Antigen presentation by thymic epithelial cells (TECs) is vital for T cell selection.
  • Previous studies indicated differences in antigen presentation between medullary TECs (mTECs) and cortical TECs (cTECs).

Purpose of the Study:

  • To investigate the biochemical basis for differential antigen presentation by cTECs and mTECs.
  • To analyze the expression and function of Major Histocompatibility Complex (MHC) class II molecules and invariant chains (Ii) in both TEC subsets.

  • Immunoprecipitation and laser confocal scanning microscopy were used to analyze MHC class II molecules and invariant chains.
  • Analysis of mRNA expression for H-2Ma and H-2Mb.
