Summary

Autoantibodies targeting the red cell anion exchanger protein band 3 were studied. While autoanti-Dib was found in 2.7% of samples, it was not the sole autoantibody present, unlike autoanti-Wrb.

Background:

  • Warm-reactive autoantibodies often target protein band 3, a red cell anion exchanger.
  • Specific epitopes on band 3 include Wrb, Dia, and Dib, which result from amino acid polymorphism.

Purpose of the Study:

  • To investigate the presence and specificity of autoantibodies against the Dib epitope on red blood cells.
  • To compare the incidence of autoanti-Dib with autoanti-Wrb.

Main Methods:

  • Testing autoantibodies from 119 patients against Di(b-) red blood cells.
  • Absorbing 74 of these autoantibodies with Di(b-) red blood cells to identify specific antibody components.

Main Results:

  • All 119 autoantibodies reacted with Di(b-) red cells, indicating no exclusive anti-Dib antibodies initially.
  • Two of 74 adsorbed autoantibodies (2.7%) contained an unadsorbed anti-Dib component.

  • The low incidence of autoanti-Dib contrasts with the higher incidence of autoanti-Wrb, despite both targeting band 3 epitopes.
