Summary

Platelet-derived growth factor-BB (PDGF-BB) and alpha 2 beta 1-integrin regulate interstitial fluid pressure (Pif). Blocking alpha 2 beta 1-integrin lowers Pif, but PDGF-BB administration normalizes it, revealing a novel role for PDGF-BB in Pif control.

Background:

  • The regulation of Pif by cellular adhesion receptors and growth factors is not fully understood.

Purpose of the Study:

  • To investigate the role of platelet-derived growth factor-BB (PDGF-BB) and alpha 2 beta 1-integrin in controlling rat skin interstitial fluid pressure (Pif).

Main Methods:

  • Interstitial fluid pressure (Pif) was measured in rat skin using glass capillaries and a servocontrolled counter-pressure system.
  • Subdermal injections of anti-alpha 2 beta 1-integrin antibodies and PDGF-BB were administered.
  • Dextran anaphylaxis was used as a model to induce changes in Pif.

  • These findings suggest PDGF-BB actively participates in regulating Pif by modulating connective tissue cell interactions with the extracellular matrix.
