Summary

Gene amplification of the SAS gene is common in osteosarcoma, particularly surface subtypes. This finding suggests SAS plays a role in osteosarcoma development and subtype differences.

Background:

  • Gene amplification, particularly of growth-promoting genes, contributes to human tumor development.
  • The SAS gene, located in the frequently altered q13-15 region of chromosome 12, is implicated in sarcomas.

Purpose of the Study:

  • To determine the frequency of SAS gene amplification in osteosarcoma.
  • To investigate the association between SAS gene amplification and osteosarcoma subtypes.

  • Southern blot analysis was employed to detect gene amplification.
  • Quantitative polymerase chain reaction (qPCR) was used for precise quantification of SAS gene amplification.

  • SAS gene amplification was observed in 36% (10 of 28) of osteosarcoma cases.
