Area of Science:

  • Psychiatry
  • Pediatric Medicine
  • Pharmacology

Background:

  • Juvenile mood disorders are increasingly recognized, yet effective medication options remain limited.
  • Nefazodone, a novel antidepressant, has not been previously studied in pediatric populations.
  • This study explores nefazodone's efficacy in treatment-refractory children and adolescents with mood disorders.

Observation:

  • Seven children and adolescents (mean age 12.4 years) with diagnosed mood disorders were treated with nefazodone.
  • Treatment duration averaged 13 weeks, with a mean daily dose of 357 mg.
  • Participants had complex conditions, often with multiple prior medication failures and comorbidities.

Findings:

  • 56% of participants showed significant improvement in depressive symptoms, as assessed by the Clinical Global Impression scale.

Related Experiment Videos

  • Two of four children with bipolar depression responded well, while two experienced mild manic activation.
  • Nefazodone was generally well-tolerated, with adverse effects reported in only three subjects.
  • Implications:

    • Nefazodone may represent a viable treatment option for pediatric mood disorders, particularly in cases resistant to other therapies.
    • The observed tolerability suggests a favorable safety profile in this population.
    • Further rigorous, controlled clinical trials are necessary to confirm these preliminary findings and establish definitive treatment guidelines.
