Summary

The metastasis-associated CD44 variant exon V6 is not expressed in cutaneous T-cell lymphomas. CD44 standard isoform (CD44H) expression does not correlate with prognosis, but CD44 V8-9 variant expression warrants further investigation in specific subtypes.

Background:

  • The lymphocyte homing receptor CD44 and its splice variants are implicated in tumor spread and poor prognosis in non-Hodgkin's lymphoma.
  • CD44 variant exon V6 expression in vitro confers metastatic potential in rat pancreatic carcinoma cell lines.

Purpose of the Study:

  • To investigate the expression of CD44 splice variants in cutaneous T-cell lymphomas (CTCL).
  • To correlate CD44 variant expression with clinicopathological subtypes and prognostic groups in CTCL.

Main Methods:

  • Immunohistochemistry was performed on CTCL samples (mycosis fungoides, Sezary syndrome, large-cell anaplastic lymphoma, HTLV1-associated cutaneous lymphoma), involved lymph nodes, and normal skin.
  • A panel of monoclonal antibodies was used, targeting CD44 standard isoform (CD44H) and variant exons V3, V6, and V8-9.
