    Summary

    Diazepam (10 mg) temporarily reduced the hypoxic ventilatory response in healthy adults, but did not affect hypercapnic response. This suggests caution in patients with respiratory issues.

    Background:

    • Diazepam is a benzodiazepine commonly used for its anxiolytic and sedative properties.
    • Understanding its impact on respiratory control is crucial for patient safety, especially in those with pre-existing respiratory conditions.

    Purpose of the Study:

    • To investigate the effects of diazepam on ventilatory control in healthy individuals.
    • To assess the specific impact of diazepam on hypoxic and hypercapnic ventilatory responses.

    Main Methods:

    • Eight healthy subjects received 10 mg of intramuscular diazepam.
    • Hypoxic and hypercapnic ventilatory responses were measured before and at various time points after diazepam administration.
    • Resting minute ventilation and end-tidal CO2 tension were also monitored.

    Main Results:

    • A significant depression of the hypoxic ventilatory response was observed at 15 and 30 minutes post-diazepam (p < 0.05).
    • No significant changes were noted in resting ventilation or end-tidal CO2 tension.
    • The hypercapnic ventilatory response remained unaffected 70 to 130 minutes after diazepam administration.

    Conclusions:

    • Diazepam can depress the hypoxic ventilatory response in normal subjects.
    • Clinicians should consider potential adverse respiratory effects in patients with compromised ventilatory function, such as chronic obstructive pulmonary disease (COPD) or acute hypoxemia.
