Summary

Chymotrypsin protease, typically found in the pancreas, was also detected in other rat organs like the stomach and spleen. This suggests a potential physiological role for this enzyme beyond its known pancreatic function.

Background:

  • Chymotrypsin-like protease is known in peripheral organs, but its synthesis outside the pancreas lacks definitive evidence.

  • To investigate the presence and synthesis of chymotrypsinogen mRNA in peripheral organs.
  • To determine if chymotrypsin-like immunoreactivity and protein exist in tissues beyond the pancreas.

  • Polymerase Chain Reaction (PCR) was used to amplify a fragment of rat pancreatic chymotrypsin mRNA.
  • In vitro transcription generated an antisense riboprobe for detecting chymotrypsin mRNA.
  • Ribonuclease protection assays, Northern blot analysis, and immunoblotting were employed to detect mRNA and protein.
