Summary

The protein isoform CncB in Drosophila selectively represses Hox protein Deformed activity. Loss of CncB causes homeotic transformations in head development, increasing segmental diversity.

Area of Science:

  • Developmental Biology
  • Genetics
  • Molecular Biology

Background:

  • The Drosophila cap 'n' collar locus encodes multiple protein isoforms.
  • Hox proteins, like Deformed (Dfd), are crucial for establishing anterior-posterior identity during embryogenesis.
  • Precise regulation of Hox gene activity is essential for correct morphogenetic outcomes.

Purpose of the Study:

  • To characterize the function of the CncB protein isoform.
  • To investigate the interaction between CncB and the Hox protein Deformed.
  • To understand CncB's role in modulating Hox-mediated cis-regulatory element activity and its impact on head development.

Main Methods:

  • Protein isoform characterization in Drosophila.
  • Analysis of gene expression patterns during embryogenesis.

Related Experiment Videos

  • Investigating cis-regulatory element activity using reporter assays.
  • Studying the effects of protein absence/reduction and ectopic expression.
  • Main Results:

    • CncB selectively represses cis-regulatory elements activated by the Hox protein Deformed.
    • CncB is expressed in specific head cells (mandibular and labral) during embryogenesis.
    • Loss or reduction of CncB leads to homeotic transformation of mandibular cells to maxillary identities.
    • This transformation is linked to persistent Deformed expression due to impaired CncB antagonism.
    • Ectopic CncB expression attenuates Deformed response elements in maxillary cells but not trunk cells.

    Conclusions:

    • CncB acts as a repressor of Deformed activity, specifically in certain head segments.
    • CncB plays a critical role in preventing homeotic transformations in the head.
    • CncB contributes to the segmental diversity of the Drosophila head by modulating Hox protein specificity.
