Insights

Bridging integrator 1 (BIN1) and Rab interactor 1 (RIN1) are downregulated in chronic myeloid leukemia (CML), impacting tyrosine kinase receptor (RTK) endocytosis. Treatment restored BIN1 and RIN1 expression, suggesting a novel therapeutic target for CML.

Area of Science:

  • Cell Biology
  • Molecular Biology
  • Hematology

Background:

  • Endocytosis regulates tyrosine kinase receptor (RTK) activity.
  • Bridging integrator 1 (BIN1) is crucial for RTK intracellular trafficking.
  • Dysregulated RTK signaling is implicated in hematological malignancies.

Purpose of the Study:

  • To investigate the role of BIN1 and its activator Rab interactor 1 (RIN1) in chronic myeloid leukemia (CML).
  • To explore the impact of these proteins on RTK endocytosis in CML.
  • To assess the effect of tyrosine kinase inhibitors (TKIs) on BIN1 and RIN1 expression in CML.

Main Methods:

  • Studied gene expression in 34 CML patients at diagnosis and 57 patients undergoing TKI treatment.
  • Performed in vitro experiments on CML and non-tumorigenic cell lines treated with Imatinib.
  • Analyzed the correlation between BIN1-RIN1 and BCR-ABL1 expression.
  • Investigated AXL receptor internalization in K562 cells.

Main Results:

  • BIN1 and RIN1 were downregulated in CML patients compared to healthy controls.
  • TKI treatment, alongside BCR-ABL1 inactivation, increased BIN1 and RIN1 expression.
  • A significant inverse correlation was observed between BIN1-RIN1 and BCR-ABL1 expression.
  • Imatinib treatment induced BIN1-RIN1 upregulation and AXL receptor internalization in K562 cells.

Conclusions:

  • BIN1 and RIN1 play a role in the altered endocytosis of RTKs in CML.
  • Restoration of BIN1 and RIN1 expression by TKIs may help normalize RTK signaling.
  • BIN1 and RIN1 represent potential therapeutic targets for managing CML by modulating RTK endocytosis.

Intracellular Signaling Affects Focal Adhesions01:17

Intracellular Signaling Affects Focal Adhesions

Integrins act both as extracellular input receivers and as intracellular processing activators. As their name suggests, integrins are entirely integrated into the membrane structure. Their hydrophobic membrane-spanning regions interact with the phospholipid bilayer's hydrophobic region. These membrane receptors provide extracellular attachment sites for effectors like hormones and growth factors. They activate intracellular response cascades when their effectors are bound and active.

Negative Regulator Molecules01:23

Negative Regulator Molecules

Positive regulators allow a cell to advance through cell cycle checkpoints. Negative regulators have an equally important role as they terminate a cell’s progression through the cell cycle—or pause it—until the cell meets specific criteria.

Receptor Downregulation in MVBs01:15

Receptor Downregulation in MVBs

Multivesicular bodies (MVBs) are mature endosomes that sort ubiquitinated proteins and then fuse with lysosomes to degrade the sorted proteins. Epidermal growth factor (EGF) and its receptor (EGFR) form a complex that can be internalized through endocytosis, sorted into an MVB, and later degraded.
The EGFR can initiate signaling pathways that  lead to cell proliferation, migration, and differentiation. Overexpression of EGFR  stimulates cells to proliferate. Excessive  EGFR...

Activation of Integrins01:15

Activation of Integrins

Integrins bind ligands and transmit information from outside the cell to inside or vice-versa through an "outside-in signaling" or "inside-out signaling."
In "outside-in signaling," external factors in the extracellular space bind to exposed ligand binding sites on integrins. This causes the inactive protein to undergo a conformational change to become active. Integrins are often clustered on the cell membrane. Repetitive and regularly spaced ligand binding...

The Intrinsic Apoptotic Pathway01:31

The Intrinsic Apoptotic Pathway

Internal cellular stress, such as cellular injury or hypoxia, triggers intrinsic apoptosis. The B-cell lymphoma 2 (Bcl-2) family of proteins are the primary regulators of the intrinsic apoptotic pathway. For example, during DNA damage, checkpoint proteins, such as Ataxia Telangiectasia Mutated (ATM protein) and Checkpoints Factor-2 (Chk2) proteins, are activated. These proteins phosphorylate p53 which further activates pro-apoptotic proteins, such as Bax, Bak, PUMA, and Noxa, and inhibits...

Diversity of Antigen Receptors01:28

Diversity of Antigen Receptors

Antigen receptors are essential components of the immune system crucial in defending the body against foreign invaders. These receptors are present on the surface of B and T cells, enabling them to recognize antigens and mount an appropriate immune response.
Before encountering any antigen, lymphocytes express these receptors. On B cells, the antigen receptor is a membrane-bound antibody molecule called BCR; on T cells, it is a T cell receptor or TCR. B and T cell receptors are composed of two...
