1Department of Clinical Laboratory and Medicine, Kyoto Prefectural University of Medicine.
Insights
Gamma delta T lymphocytes, crucial in immunology, were quantified in 151 healthy adults. Their numbers slightly decrease with age but show no significant sex-based differences.
Area of Science:
- Immunology
- Cell Biology
- Flow Cytometry
Context:
- Gamma delta T lymphocytes are implicated in various immunological disorders.
- Clinical need exists to establish normal ranges for these cells in peripheral blood.
- Understanding normal cell populations is vital for diagnosing immune system abnormalities.
Purpose:
- To determine the normal population of gamma delta T lymphocytes in healthy adults.
- To establish reference ranges for cell counts across different age groups.
- To investigate potential correlations between cell numbers and demographic factors like age and sex.
Summary:
- 151 healthy volunteers aged 20-69 years were analyzed using dual-color flow cytometry.
- Gamma delta T lymphocytes constituted 3.8 +/- 2.5% of peripheral blood lymphocytes (range: 0.6-16.1%).
- A significant inverse correlation was observed between gamma delta T cell counts and increasing age, with no significant sex-based differences.
Impact:
- Provides essential reference values for gamma delta T lymphocyte populations in healthy individuals.
- Aids in the diagnosis and monitoring of immunological disorders.
- Contributes to a deeper understanding of T cell subset dynamics throughout the lifespan.
Abstract:
Since gamma delta T lymphocytes has been found to be increased in patients with some immunological disorders, there exists clinical needs to determine the population of the cells in peripheral blood in normal subjects. We counted the number of gamma delta T lymphocytes in samples obtained from 151 healthy volunteers, aged from 20 to 69 years, by using a dual-color flow cytometry with fluorescence-conjugated CD3 and anti-TCR-gamma delta antibodies. Distribution ratio of gamma delta T lymphocytes among peripheral blood lymphocytes was 3.8 +/- 2.5% (range, 0.6-16.1%). Difference in the ratio and the number of gamma delta T lymphocytes among sex was not significant, but those decreased slightly but significantly with age.
