Enzymes are proteins made of amino acids. The functional group of each constituent amino acid catalyzes a wide variety of chemical reactions via ionic interactions or acid-base reactions. Alkylation of β-Ketoester Enolates: Acetoacetic Ester Synthesis

Acetoacetic ester synthesis is a method to obtain ketones from alkyl halides and β-keto esters. The reaction occurs in the presence of an alkoxide base that abstracts the acidic proton of the β-keto esters. The step results in an enolate ion which is doubly stabilized. The enolate then reacts with an alkyl halide via the SN2 process to produce an alkylated ester intermediate with a new C–C bond. The hydrolysis of the intermediate, followed by acidification, results in an alkylated β-keto acid.

Sus antecedentes:

  • Las sintetasas policetidas (PKS) son enzimas cruciales que construyen moléculas policetidas a través de reacciones de condensación secuenciales.
  • El dominio beta-cetoacil sintasa (KS) es el núcleo catalítico responsable del paso de formación de enlaces C-C en el ensamblaje PKS.
  • La comprensión de los mecanismos de dominio de KS es clave para la ingeniería de PKS para la síntesis de compuestos novedosos.

Objetivo del estudio:

  • Investigar el mecanismo catalítico de dos dominios de KS altamente homólogos, NonJ y NonK, de la vía de biosíntesis de la nonactina.
  • Para determinar si el NonJ y el NonK exhiben formación de enlaces C-C canónicos o una actividad catalítica alternativa.
  • Explorar el potencial de estos dominios de KS para la ingeniería de PKS y la expansión de la diversidad de policetidos.

Principales métodos:

  • Caracterización genética del grupo de genes de la biosíntesis de la nonactina en Streptomyces griseus.
  • Ensayos bioquímicos para analizar la actividad de los dominios de KS No-J y No-K.
  • Análisis comparativo de NonJ y NonK con dominios conocidos de KS.

Principales resultados:

  • Se identificaron y caracterizaron los dominios NonJ y NonK KS de Streptomyces griseus.
  • Estos dominios KS catalizan la condensación de sustratos de la acilcoenzima A (CoA) mediante la formación de enlaces C-O, que se desvían de la formación típica de enlaces C-C.
  • Los dominios KS identificados muestran una alta homología con los KS conocidos, lo que sugiere una divergencia en la función catalítica dentro de la familia KS.

Conclusiones:

  • El descubrimiento de NonJ y NonK revela una nueva actividad catalítica dentro de los dominios PKS KS, formando enlaces C-O en lugar de C-C.
  • Esta química única ofrece nuevas posibilidades para la ingeniería de PKS.
  • Aprovechar esta capacidad de formación de enlaces C-O puede ampliar significativamente el alcance y la diversidad de la biosíntesis de policeturo, lo que lleva a nuevos compuestos.
