Sus antecedentes:

  • Los elementos no codificantes altamente conservados (HCNEs) se encuentran con frecuencia cerca de genes críticos para el desarrollo.
  • Se supone que estas regiones ricas en HCNE albergan elementos regulatorios esenciales que controlan los procesos de desarrollo.

Objetivo del estudio:

  • Para investigar el paisaje epigenético de las regiones ricas en HCNE en células madre embrionarias (ES) de ratón.
  • Identificar patrones específicos de modificación de histonas asociados con estas regiones reguladoras y sus genes vinculados.

Principales métodos:

  • Análisis de los patrones de metilación de histonas (H3K27me y H3K4me) a través de 56 grandes loci ricos en HCNE en células ES de ratón.
  • Correlación de los patrones de metilación con la expresión de los genes cercanos del factor de transcripción (TF).

Principales resultados:

  • Identificación de "dominios bivalentes" caracterizados por la metilación de H3K27 que abarca la metilación de H3K4 en las células ES.
  • Los dominios bivalentes se encontraron predominantemente en genes de factores de transcripción importantes para el desarrollo expresados en niveles bajos.
  • Se observó una fuerte correlación entre la secuencia de ADN y la metilación de histonas en las células ES, disminuyendo con la diferenciación.

Conclusiones:

  • Los dominios bivalentes pueden funcionar para silenciar los genes de desarrollo en las células ES, manteniéndolos en un estado de equilibrio para una activación rápida.
  • La secuencia de ADN juega un papel crucial en el establecimiento del estado epigenético inicial de estas regiones.
  • Se sugiere un nuevo mecanismo basado en la cromatina para mantener la pluripotencia, que implica la interacción de la secuencia de ADN y las modificaciones epigenéticas.
